Herein we report a novel portable platform for the rapid Vitamin D3 (25-OHD3) detection in untreated serum samples. The electrochemical immunosensor was developed by immobilizing the 25-OHD3 antibody (Ab 25-OHD) on a graphite screen printed electrode by means of glutaraldehyde cross-linking on an amino modified surface. The Ab immobilization technique was optimized by monitoring the protein density by Surface Plasmon Resonance experiments. The influence of pH and interfering species was evaluated, and the optimized platform was calibrated by using Differential pulse voltammetry in different antigen standard solutions. The immunosensor was then tested for human serum analysis and its results were compared with the chemiluminescence-immunoassay (CLIA) reference method with a good agreement. These features, makes our platform a valuable tool for POCT development.
