In order to clarify the mechanism(s) of anthracycline activation and the subsequent effects on red blood cells, we have made a series of experiments using daunomycin, which has been claimed to cause damage in drug-loaded erythrocytes. We here report preliminary results from this study, during which the ESR signal of the daunomycin radical generated by intact red blood cells and the related production of O2- were seen for the first time. The results suggest that the radical we observed is formed on the outside of the erythrocyte; no major damage to cell function and integrity was observed.
