A reversed-phase HPLC method for the microdetn. of CN- is based on its precolumn derivatization according to W.J. Koenig (1904) to produce a polymethine dye and spectrophotometric or fluorometric detection. Hypersil ODS or μ-Bondapack C18 can be used as the stationary phase. The mobile phase was a mixt. of 50/50 H2O-MeOH and 0.010 M NH4H2PO4 (pH = 4.7). The calibration curve was linear in the range 96-11,500 and 37-769 fmol for the spectrophotometric and fluorometric detection, resp.,; the detection limit was 95 and 7 fmol, resp. The coeff. of variation was 2%. The method can also be used to det. SCN-. [on SciFinder(R)]
