Site-directed mutagenesis techniques have been applied to the glyA gene of E. coli coding for serine hydroxymethyltransferase, in order to substitute amino acid residues known to be at the active site of the enzyme. Two high efficient mutagenesis methods have been used that specifically select, in vivo or in vitro, mutant DNA with respect to the wild-type one.
