In the Crabtree-negative yeast Kluyveromyces lactis respiratory and fermentative metabolism coexist. Pyruvate, which is the branching point between these two metabolic pathways, is decarboxylated by the Pyruvate decarboxylase (PDC) or the Pyruvate dehydrogenase (PDH) enzymes. In K. lactis respiration and fermentation are connected by the pyruvate bypass and the reciprocal acetyl-CoA exchange between cytoplasm and mitochondria, which allows the growth on glucose of both PDC and PDH mutant strains. The PDC gene of K. lactis (KlPDC1) is regulated at the transcriptional level by carbon sources, by oxygen and by several gene products involved either in general regulation or in glycolysis. Metabolic engineering of strains defective in pyruvate utilization allows the construction of strains producing heterologous metabolites with high yield as unique final fermentation product.
